The Salentini dismissed Di Francesco in the wake of Saturday's 4-2 defeat at Napoli that left the team bottom of the Serie A standings.
Lecce, who have claimed one victory in their last 11 games, confirmed in a club statement: "Lecce announces it has parted company with Eusebio Di Francesco.
"The club also announces it has reached an agreement with Serse Cosmi to coach the team.''
The Cosmi, 53, had been out of a job since being fired by Palermo earlier this year after less than three months in charge of the Sicilian outfit.
Cosmi's first game in charge will be against Lazio on Saturday.
