Arsenal left-back Kieran Gibbs has been called up as his replacement.
Cole was substituted with 15 minutes remaining of Chelsea's 3-1 win at Norwich on Sunday, replaced for tactical reasons with Eden Hazard by manager Jose Mourinho as the Blues pushed for the win.
However, the defender later complained of a problem with his ribs and underwent a scan on Monday.
Cole did not report with the rest of Roy Hodgson's squad at St George's Park, opting to stay at the club for further treatment, meaning he will miss the first of England’s two crucial Group H qualifying matches at Wembley on Friday.
It has not been confirmed whether Cole will miss Tuesday’s game against Poland, also at Wembley.
