Stam, 40, has agreed a three-year contract and will link up with the Dutch giants in July.
He will work under former international colleagues Frank de Boer and Dennis Bergkamp, Ajax's manager and assistant manager respectively.
Stam, who won 47 caps for Netherlands, called time on his playing career in 2007 after a brief spell with Ajax. He had earlier represented clubs including PSV Eindhoven, Manchester United, Lazio and AC Milan.
After hanging up his boots, the former centre-back had worked for United as a scout.
