Keane, 32, was unable to play a full part in training in Malahide, and Republic coach Giovanni Trapattoni has decided that he will not play at the Aviva Stadium.
However, it is hoped that he will have recovered in time to play in Tuesday's game against Faroe Islands.
On its official Twitter feed, the Football Association of Ireland confirmed: "Robbie Keane has been ruled out of the Germany game with an Achilles injury. Will hopefully be fit for Faroes."
Trapattoni will hand Jon Walters a lone striking role for Friday's game in the abscence of Keane.
Ireland team to play Germany: Westwood; Coleman, O'Shea, O'Dea, Ward; McGeady, Andrews, McCarthy, Fahey, Cox; Walters.
